Yes

Dot 3, 4 & 5.1 are water absorbing and have 12 different basic formulas of glycol & ether.

Dot 5 is silicone based, is not water absorbing and has 3 different basic formulas.

While it’s permissible to mix DOT 3, DOT 4 and DOT 5.1 brake fluids, DOT 5 Silicone fluid cannot be mixed with any other type. If the system is not completely purged before conversion, the mixture may gel and result in very poor braking.

I have run a lot of vehicles on DOT5. It's always easier when replacing parts, no need to flush them. So if going with a new caliper, no issue. If a used caliper, just take it apart and clean everything. Use some DOT5 to assemble it as well. As for flushing, alcohol is the material to use. But all you need to do is disassemble and clean.

George, I am not an expert on this...just know the basics.
If a new caliper, I would expect dot 5 silicone brake fluid to not be an issue.
If used, then flush it well, as PetesPonies already suggested.
