Should be a straight swap especially if you stick to the same year. Spindles changed one year but can't remember when

08 changed to 25mm (from the 3/4" every 2000-2007 uses). The bearing cup got shallower for the thinner bearings.
An 08-12 (maybe... whatever year the Iron got the new 13 spoke with the tapered hub) Sporty with the 13 spoke cast narrow wheel should work fine though. I'm pretty sure they were just running a spacer to fill the extra 6mm in those as opposed to redesigning the hub for an existing mold. So anything pre-08 works basically.
And you can re-use your rotor if it's still good. No issues there. The hub is the same width on all of these wheels, and the rotor spacing hasn't changed since they went to sealed bearings.

2005-2007 use a 1" (25.4mm) axle AFAIK. Most any single disk brake sportster wheel should work, just need to match bearings, axles, spacers. That link has a lot of information and should be a few threads on here for other situations. I think some other Harley models wheels could fit IIRC.

Depends on whether you are referencing the front or rear wheel
Front:
2000-2007 - 3/4” Axle
Rear:
2000-2004 - 3/4“ Axle
2005-2007 - 1” Axle
From the Sportsterpedia, check under wheels, brakes & tires

Yeah, I was thinking front the whole time... Rear is different, true.
You can get 15mm thick 1” bearings though, so not as restrictive in wheel selection back there if you‘ve got a 1” axle and need them.
But no, newer 25mm wheels do not work on either end with original bearings. The hubs are spaced for 15mm thick bearings in the new wheels and the 3/4” and 1” bearings are 21mm thick. You get overhang. Not sure how safe that would be even if you made spacers fit the assembly.
